ABSTRACT To investigate the interaction between two major ear-rotting pathogens, maize ears were inoculated with either Fusarium graminearum, F. moniliforme, or an equal mixture of the two. Silk and kernel tissues were periodically harvested throughout the growing season so that a time course of the experimental variables (disease severity, ergosterol content, fungal DNA content, and mycotoxin concentration) could be recorded. Over the 3 years tested (1992 to 1994), the highest levels of disease and ergosterol were found in the F. graminearum treatment, followed by the mixture treatment (F. graminearum plus F. moniliforme) and, finally, the F. moniliforme treatment. Kernel ergosterol content and disease rating were correlated for both pathogens, but the highest correlation coefficients were obtained in the F. graminearum treatment. The DNA analysis revealed that, in the mixed inoculum, F. moniliforme had a greater growth rate than did F. graminearum. In 1994, appreciable F. moniliforme from natural inoculum was found in the F. graminearum treatment. Fumonisin B(1) levels did not differ between the F. moniliforme treatment and the mixed inoculum treatment. The effect of temperature on the growth rate of the two species explained some of the field results, with temperatures in the silks being more favorable to F. moniliforme. Data on the growth rate on silks obtained by the incorporation of radiolabeled precursor to ergosterol demonstrated that F. graminearum was able to grow well at 26 to 28 degrees C, whereas F. moniliforme grew well over a broader range, including at higher temperatures.  相似文献

The purpose of this study was to investigate magnetic resonance imaging of the normal canine and feline eye, orbit and optic nerves using proton density-weighted, T1-weighted and T2-weighted images. The clinical application of magnetic resonance imaging in veterinary ophthalmology was also investigated using three clinical cases: a feline orbital melanoma, a feline optic nerve meningioma, and a canine orbital fibrosarcoma. Gadolinium diethylenetriamine pentaacetic acid enhanced magnetic resonance imaging was completed on the case of feline optic nerve meningioma. Magnetic resonance imaging provides excellent anatomical detail of the canine and feline eye, orbit, and optic nerves due to its superior soft tissue contrast, and its multiplanar and multislice imaging capability. Therefore it is of value for diagnostic imaging of some ophthalmic and neuro-ophthalmic conditions in the dog and cat.  相似文献

Two trials were conducted to determine the effect of cambendazole against natural infections of the swine kidneyworm, Stephanurus dentatus, in sows. In the first trial, nine sows were given a single dose of 20 mg cambendazole/kg body weight, in the feed, and eight sows were not treated. In the second trial, five sows were given 40 mg cambendazole/kg body weight and four were untreated. Urine volume, S. dentatus eggs per milliliter of urine, egg hatchability, and larval survival to the third stage were determined pre and post treatment. Adult kidneyworms were counted at necropsy, 14 days after treatments. Only a temporary decline in egg hatchability was observed on the day after treatment in each trial, and cambendazole was not found to be effective against adults of S. dentatus.  相似文献

The purpose of this study was to investigate total baseline plasma cortisol and adrenocorticotropic hormone (ACTH) concentrations, and ACTH-stimulated cortisol concentrations in foals from birth to 12 wk of age. Plasma (baseline) cortisol and ACTH concentrations were measured in 13 healthy foals at birth and at 1, 2, 3, 4, 5, 7, 10, 14, 21, 28, 42, 56, and 84 d of age. Each foal received cosyntropin (0.1 μg/kg) intravenously. Plasma cortisol concentrations were measured before (baseline), and 30, and 60 min after cosyntropin administration at birth and at 3, 5, 7, 10, 14, 21, 28, 42, 56, and 84 d of age. Compared with baseline, cortisol concentration increased significantly 30 min after administration of cosyntropin on all days. Cortisol concentration was highest at birth, measured at 30 and 60 min after cosyntropin administration, compared with all other days. With the exception of birth measurements, cortisol concentration was significantly higher on day 84, measured at 30 and 60 min after cosyntropin administration, when compared with all other days. Baseline plasma ACTH was lowest at birth when compared with concentrations on days 2, 3, 4, 5, 7, 10, 14, 42, 56, and 84. Administration of 0.1 μg/kg of cosyntropin, IV, reliably induces cortisol secretion in healthy foals. Differences in the magnitude of response to cosyntropin are observed depending on the age of the foal. These data should serve as a reference for the ACTH stimulation test in foals and should be useful in subsequent studies to evaluate the hypothalamic-pituitary-adrenal axis in healthy and critically ill foals.  相似文献

Mastitis is an economically important disease and its subclinical state is difficult to diagnose, which makes mitigation more challenging. The objectives of this study were to screen clinically healthy ewes in order to 1) identify cultivable microbial species in milk, 2) evaluate somatic cell count (SCC) thresholds associated with intramammary infection, and 3) estimate relationships between udder and teat morphometric traits, SCC, and ewe productivity. Milk was collected from two flocks in early (<5 d) and peak (30 to 45 d) lactation to quantify SCC (n = 530) and numerate cultivable microbial species by culture-based isolation followed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS; n = 243) identification. Within flock and lactation stage, 11% to 74% (mean = 36%) of samples were culture positive. More than 50 unique identifications were classified by MALDI-TOF MS analysis, and Bacillus licheniformis (18% to 27%), Micrococcus flavus (25%), Bacillus amyloliquefaciens (7% to 18%), and Staphylococcus epidermidis (26%) were among the most common within flock and across lactation stage. Optimum SCC thresholds to identify culture-positive samples ranged from 175 × 103 to 1,675 × 103 cells/mL. Ewe productivity was assessed as total 120-d adjusted litter weight (LW120) and analyzed within flock with breed, parity, year, and the linear covariate of log10 SCC (LSCC) at early or peak lactation. Although dependent on lactation stage and year, each 1-unit increase in LSCC (e.g., an increase in SCC from 100 × 103 to 1,000 × 103 cells/mL) was predicted to decrease LW120 between 9.5 and 16.1 kg when significant. Udder and teat traits included udder circumference, teat length, teat placement, and degree of separation of the udder halves. Correlations between traits were generally low to moderate within and across lactation stage and most were not consistently predictive of ewe LSCC. Overall, the frequencies of bacteria-positive milk samples indicated that subclinical mastitis (SCM) is common in these flocks and can impact ewe productivity. Therefore, future research is warranted to investigate pathways and timing of microbial invasion, genomic regions associated with susceptibility, and husbandry to mitigate the impact of SCM in extensively managed ewes.  相似文献

An 11-year-old male intact Staffordshire Bull terrier was referred for diabetic ketoacidosis. Abdominal ultrasonographic examination revealed a 5 cm × 2 cm intraluminal caecal mass-like structure. Exploratory laparotomy and typhlectomy were subsequently performed. Histopathology of the caecal mass-like structure was consistent with a caecal mucocoele, defined as a cystic dilation of the caecal lumen with stasis of mucus. This lesion has been previously described in humans, where it is termed an appendiceal mucocoele. The patient was euthanased 58 days post-operatively due to unrelated diabetic complications.  相似文献

Net mineralization of sulfur and nitrogen was studied in three Canadian Prairie soils using two commonly used incubation methods. In the open system technique, where the soils were leached periodically II.3–11.8 μ g SO2?4 -S g?1 soil was mineralized in 17 weeks. Little mineralization or a net immobilization of sulfur (from 1.4 to 1.3 μ g SO2?4-S g?1 soil) was observed in a closed system where the soils were left undisturbed throughout incubation. Changes in the specific activity of 35S-labelled soil solution sulfate during the closed incubation indicated that mineralization-immobilization processes were occurring simultaneously resulting in minimal net changes in CaCl2-extractable SO2?4 concentrations. The amounts of mineralized nitrogen (32.6–57.8 μg N g?1 soil) were found to be independent of the incubation method employed.  相似文献
